Delivery
Getting a nuclease-resistant siRNA into the right cells is the central challenge of the modality. Two approaches dominate the clinic.
- Conjugate — a targeting ligand (e.g. GalNAc) covalently attached to the siRNA.
- Lipid nanoparticles — multicomponent lipid formulations that encapsulate the payload.
